Während eines langen Sandsturms geht der Kontakt zur ersten menschlichen Kolonie auf dem Mars verloren. Satellitenbilder zeigen, dass die Anlage stark beschädigt ist, und die vierundfünfzig Kolonisten, die Colony One ihr Zuhause nannten, gelten als tot. Drei Jahre später landet eine neue Mission auf der Oberfläche des Planeten, um zu untersuchen, was von der verlassenen Anlage übrig geblieben ist.
Schnell stellt die Crew fest, dass die Kolonie doch nicht so tot ist, wie alle dachten. Irgendjemand ist noch am Leben - versteckt sich aber. Doch während sie nach dem wichtigen Zeugen sucht, wird die Besatzung von einer seltsamen Krankheit befallen. Die Biologin Dr. Jann Malbec muss so schnell wie möglich den Ursprung der Krankheit und einen Weg zu ihrer Bekämpfung finden.
Bei ihren Nachforschungen stößt Jann auf ein dunkles und tödliches Geheimnis, das in Colony One lauert. Ein Geheimnis, das nicht nur die Besatzung, sondern auch die gesamte Bevölkerung der Erde bedroht. Mit begrenzten Ressourcen und unter Zeitdruck muss die Biologin Antworten auf Fragen finden, die besser nie gestellt worden wären. Denn wenn sie scheitert, wird keiner von ihnen nach Hause zurückkehren.
